5.Evaluation of diversity of bacterial population on rhizoplane by PCR-fingerprinting

Enterobacterial repetitive intergenic consensus (ERIC) sequences and restriction enzyme fragments of 16S rDNA from Bradyrhizobium strains on soybean roots were analysed by polyacrylamide gel electrophoresis. Both ERIC sequences and 16S rDNA were amplified from more than 106 cells of bradyrhizobia by polymerase chain reaction to be visualized on the electrophoresis gels. Analyses of restriction enzyme fragments of 16S rDNA have enabled distinction among different species of Bradyrhizobium. Isolated strains were distinguishable in terms of molecular sizes of amplified ERIC sequences on the electrophoresis gels.
